Background:
Granulocyte colony stimulating factor receptor (G-CSFR) exists on the cell surface of the hematopoietic progenitor cell in bone marrow, in mature hematopoietic cells and stromal cells. Binding with granulocyte colony stimulating factor (G-CSF) to the extracellular ligand-binding region of G-CSFR triggers G-CSFR to form homodimer. The conformational changes in G-CSFR activate the cytoplasmic portion of the receptor and trigger signal transduction through Jak Lyn, Stat and erk1/2. The pathway is important for hematopoiesis and was found to be implicated in congenital neutropenia, myelodysplastic syndrome and acute myeloid leukemia.
Applications:
Western Blot: The antibody, when used at concentration of 1-10mg/mL will allow visualization of 250ng/lane of human G-CSFR-MBP.
The antibody recognizes denatured G-CSFR in Western blot following SDS-PAGE. It is suggested to use 200mg/lane of cell lysate or immunoprecipitated protein.
